PTPN22 may play a role in regulating the function of Cbl and its associated protein kinases. PTPN22 also acts as a negative regulator of T-cell receptor (TCR) signaling. PTPN22 dephosphorylates and inactivates the SRC family kinases. Defects in PTPN22 are a cause of susceptibility to systemic lupus erythematosus (SLE).
