Background: SMAD2 is a transcriptional modulator activated by TGF-beta and activin type 1 receptor kinase. SMAD2 is a receptor-regulated SMAD (R-SMAD). SMAD2 is phosphorylated by the activated receptor kinase upon ligand stimulation, and forms a complex with other SMADs. The SMAD complex then translocates into the nucleus to regulate transcription. SMAD2 may act as a tumor suppressor in colorectal carcinoma.
